Axioscan 7 troubleshooting

  • Increase the number of support point
    • Click on “Adapt Selected Profil”
    • Step 6 “Fine Focus”
    • In “Onion Skin”, increase the parameter at 0.2
    • Click on “Finish” 
  • Redo the Autofocus (AF)
    • Click on “Adapt Selected Profil”
    • In Step 5 then Step 6, click on “Live”, then click on “AF”
    • Click on “Finish”
    • If it doesn’t work, increase the range of the Z-stack at the step 6 and redo the autofocus
  • The slice might be too thick, please check with a member of the facility.
  • Define your channel of interest as reference for the focus point in Step 6 “Fine Focus”, by right-clicking on the channel of interest and clicking on “Define as reference”
  • Change the detection thershold
    • Click on “Check Profile”
    • Change the threshold by moving the white boundary of the histogram
  • If the automatic detection is impossible, switch to manual mode and draw around your slices using the ZEN drawing tools, then click on “Sort” to define the order of acquisition
  • Change the reference channel used for stitching
    • Click on “Adapt Selected Profile”
    • In Step 7, right click on another channel and define it as reference
  • Click on “New” to change the profile and start from zero
  • Click on “Preview” to adapt and modify the previous profile
